False positive DAT results are most often associated with the use of refrigerated, clotted blood samples in which complement components coat RBCs in vitro. Any positive DAT result obtained from a clotted blood sample should be confirmed using a freshly collected EDTA-anticoagulated specimen.

Cells with a positive DAT due to complement alone will normally yield an eluate containing no antibody activity.
